I'm trying to standardise the open field test using Diazepam. I'm having some trouble with it and was hoping to get inputs.

I used 3 doses of diazepam, 0.3, 0.5 and 1mg/kg at a dose volume of 5ml/kg. With the 1mg dose, animals are initially fine. However, when I test them in the open field after 30min, the locomotion activity is poor. At the 0.3mg dose also, animals are anxious and do not explore. I have had some success with the 0.5mg dose but it does not compare to published literature.

1. In your experience, what doses of diazepam work well for rats ( I am using SD rats, 350 - 370g)
2. How long after diazepam injection should I do behavioural testing (30 minutes or 60min). What decides this time frame?
3. What are some behavioural patterns you see in rats immediately after diazepam injection
